Vector unpacking as a potential barrier for receptor-mediated polyplex gene delivery.
Ligand-conjugated polymer (polyplex) gene delivery vectors have strong potential as targeted, in vivo gene transfer vehicles; however, they are currently limited by low delivery efficiency. A number of barriers to polyplex-mediated delivery have been previously identified, including receptor binding, internalization, endosomal escape, and nuclear localization. However, based on understanding of...
متن کاملTargeting hepatocytes for drug and gene delivery: emerging novel approaches and applications.
The asialoglycoprotein receptor (ASGP-R) on mammalian hepatocytes provides a unique means for the development of liver-specific carriers, such as liposomes, recombinant lipoproteins, and polymers for drug or gene delivery to the liver, especially to hepatocytes. The abundant receptors on the cells specifically recognize ligands with terminal galactose or N-acetylgalactosamine residues, and endo...

Overexpression of the hepatocyte growth factor receptor/c-Met proto oncogene on the surface of a variety of tumor cells gives an opportunity to specifically target cancerous tissues. Herein, we report the first use of c-Met as receptor for non-viral tumor-targeted gene delivery. Sequence-defined oligomers comprising the c-Met binding peptide ligand cMBP2 for targeting, a monodisperse polyethyle...
متن کاملCucurbituril-based nanoparticles: a new efficient vehicle for targeted intracellular delivery of hydrophobic drugsw
Cucurbituril-based nanoparticles (CB[6]NPs) serve as new efficient vehicles for delivery of hydrophobic drugs, which have unique features including (1) a high drug loading capacity and efficiency, (2) noncovalently tunable surfaces, (3) efficient delivery of hydrophobic drugs into a cancer cell by receptor-mediated endocytosis, and (4) facile release of drugs into cytoplasm, which enhances the ...
متن کاملHepatocyte-targeting gene transfer mediated by galactosylated poly(ethylene glycol)-graft-polyethylenimine derivative
Biscarbamate cross-linked polyethylenimine derivative (PEI-Et) has been reported as a novel nonviral vector for efficient and safe gene transfer in our previous work. However, it had no cell-specificity. To achieve specific delivery of genes to hepatocytes, galactosylated poly(ethylene glycol)-graft-polyethylenimine derivative (GPE) was prepared through modification of PEI-Et with poly(ethylene...
